Expression oatten of PDLs17, PDL cell-specific protein, on the tooth periodontium of developing mice.

Abstract
PDLs17. PDL-specific gene, had previousely identified the cDNA for a novel protein from cultured PDL fibroblasts using subtraction hybridization between gingival fibroblasts and PDL fibroblasts. The purpose of this study was to observe the immunohistochemical localization of PDLs17 on the developing periodontium of mice.
Developing mice (post-natal mice; P4, P10, P15) were recovered and decapitated, and the heads were fixed overnight at in a freshly prepared solution of 4% parformaldehyde. Some specimens were decalcified for 2-4 weeks in a solution containing 10% of the disodium salt of ethylenediamine-tetraacetic acid (EDTA). Next, tissues were dehydrated, embedded in paraffin and sectioned serially at 6§­ in thickness. Polyclonal antiserum raised against PDLs17 peptides, SEAVHETDLHDGC, were made, and immunostained the tooth, periodontium, developing bone and bone marrow of developing mice.

The results were as follows.

1. Immunohistochemical analysis showed that PDLs17 is preferentially expressed in the developing tooth follicle, but not in tooth pulp and overlying gingival mucosa of 4-day-old developing mouse. PDLs17 also expressed in preosteoblast-like cells in the developing alveolar bone of 4-day-old mouse.

2. PDLs17 expressions were observed in developing tooth follicle, tooth pulp and alvolar bone of 10-day-old developing mouse.

3. PDLs17 protein were observed in PDL cells, tooth pulp and surrounding alveolar bone of 15-day-old tooth-eruping mouse. However, there were no PDLs17 expression in ameloblasts and odontoblasts.

These results suggestd that PDLs17 might expressed at the initial stage of diffirentiation when the undifferentiated mesenchymal cells in the bone marrow and PDL differentiation from tooth follicle. However, more research needs to be performed to gain a better understanding of the exact function of PDLs17 during the differentiation of tooth pulp cells and PDL cells.
